The success of extracellular vesicles (EVs) in diagnostics and therapeutics depends on scalable, automated, reproducible isolation methods that consistently produce high recovery of pure EVs. The most common methods for EV isolation are burdensome, difficult to automate, and difficult to scale, requiring complex manual processes. Furthermore, these methods exploit a single physical property, such as density or size, to isolate EVs, forcing a tradeoff between recovery and purity. We introduce dielectrophoresis, a new chip-based EV isolation technique that exploits the dipole moment of EVs, which is defined by two physical properties at once—size and mobile charge. This technique provides an automated, scalable solution to reproducibly deliver high yields of pure EVs.
